In the Commonwealth of Kentucky, Interstate 75 runs from near Williamsburg to Covington by way of Lexington. Interstate 75 enters the Cumberland Plateau region from Tennessee, then descends into the Bluegrass region through the Pottsville Escarpment before crossing the Ohio River into Ohio.

Interstate 75 follows along the U.S. Route 25 corridor for the entire length of Kentucky.

*From milepost 156 to milepost 139 near Corinth (2 lanes each way, currently in the process of being widened to 3 lanes each way)
*From milepost 139 to milepost 70 south of Berea (3 lanes each way)
*From milepost 70 to milepost 56 (2 lanes each way)
*From milepost 52 to milepost 56 (4 NB - extra climbing lane, 3 SB)
*From milepost 52 to milepost 41 north of London (2 lanes each way, currently in the process of being widened to 3 lanes each way)
*From milepost 41 to milepost 38 bypassing London (3 through lanes each way, NB auxiliary lane for KY 80 exit and SB auxiliary lane for KY 192 exit)
